Il dominio chinasi FAK purificato e attivato (410–689 aa) viene fatto reagire con 50 μM di ATP e 10 μg per pozzetto di un polimero peptidico casuale di Glu e Tyr, p(Glu/Tyr), in tampone di chinasi [50 mM HEPES (pH 7,5), 125 mM NaCl e 48 mM MgCl2] per 15 min. La fosforilazione di p(Glu/Tyr) viene sfidata con il composto diluito in serie a concentrazioni 1/2-Log a partire da una concentrazione massima di 1 μM. Ogni concentrazione viene testata in triplicato. La fosforilazione di p(Glu/Tyr) viene rilevata con un anticorpo generale anti-fosfotirosina (PY20) seguito da un anticorpo IgG di capra anti-topo coniugato con perossidasi di rafano (HRP). Viene aggiunto il substrato HRP e le letture di assorbanza a 450 nm vengono ottenute dopo l'aggiunta della soluzione di arresto (2 mol/L H2SO4).
