Panobinostat (LBH589) induce l'apoptosi tra le cellule MOLT-4 e Reh in modo tempo- e dose-dipendente, ed è più potente nelle MOLT-4 che nelle cellule Reh. Previene marcatamente la crescita di entrambe le linee cellulari in modo dose-dipendente a 48 ore e provoca un aumento di 2-3 volte del numero di cellule nella fase G2/M del ciclo cellulare rispetto alle cellule di controllo. Questo composto è associato all'induzione dell'acetilazione dell'istone H3K9 e dell'istone H4K8, nonché alla diminuzione dei livelli di espressione di c-Myc in modo dose-dipendente. Aumenta anche i livelli di espressione di p21 e diminuisce i livelli di c-Myc dopo un aumento iniziale alla dose più bassa (10 nM) nelle cellule Reh. Inoltre, porta a un aumento sostanziale dei livelli di mRNA dei geni proapoptotici e di riparazione del DNA e induce un aumento dei livelli di istoni H3 e H4 acetilati al promotore GADD45G. Inoltre, inibisce la crescita di linee cellulari di cancro del polmone non a piccole cellule (come H1299, L55 e A549 umane con IC50 di 5 nM, 11 nM e 30 nM, rispettivamente), mesotelioma (come OK-6 e Ok-5 umane con IC50 di 5 nM e 7 nM, rispettivamente) e linee cellulari di cancro del polmone a piccole cellule (come RG-1 e LD-T umane con IC50 di 4 nM e 5 nM, rispettivamente).
In vivo
Nei modelli animali di cancro del polmone e mesotelioma, Panobinostat (LBH589) diminuisce marcatamente la crescita tumorale del 62%. È ugualmente efficace in topi immunocompetenti e con immunodeficienza combinata grave, suggerendo che l'inibizione della crescita tumorale da parte di questo composto non è dovuta a effetti immunologici diretti. La somministrazione giornaliera, somministrata per via intraperitoneale a 20 mg/kg per 5 giorni alla settimana, porta a una diminuzione media della crescita del 70%. Rispetto ai tumori di controllo corrispondenti, si traduce in una diminuzione del 53% per i tumori derivati da H526, una diminuzione dell'81% per i tumori derivati da BK-T, una diminuzione del 76% per i tumori derivati da RG-1 e una diminuzione del 70% per i tumori derivati da H69. A differenza della mancanza di regressione tumorale osservata nei tumori xenotrapiantati derivati da NSCLC e Meso trattati in condizioni e dosi identiche, LBH589 porta a una drammatica regressione tumorale nei tumori derivati da SCLC e nei tumori derivati da RG-1.
