Flavopiridol (Alvocidib) displays less activity against unrelated kinases such as MAP, PAK, PKC, and EGFR with IC50 of >14 μM. It significantly inhibits the colony growth of HCT116, A2780, PC3, and Mia PaCa-2 cells with IC50 of 13 nM, 15 nM, 10 nM and 36 nM, respecitively. This compound also potently inhibits the activity of Glycogen synthase kinase-3 (GSK-3) with an IC50 of 280 nm. Compared with other CDKs, it inhibits the activity of CDK7 less potently with IC50 of 875 nM. At 0.5 μM, it inhibits both pSer807/811 Rb and pThr199 NPM, whereas mild changes are observed at pThr821 Rb. It also decreases the overall RNA polymerase II level, as well as the phosphorylation of RNA polymerase II on the CTD repeats at Ser2 Ser5. As a broad spectrum CDK inhibitor, it can inhibit cell cycle progression in either G1 or G2. At 0.3 μM, it induces G1 arrest in either MCF-7 or MDA-MB-468 cells by inhibition of the CDK4 or CDK2 kinase activity. It exhibits potent cytotoxicity against a wide variety of tumor cell lines with IC50 values ranging form 16 nM for LNCAP to 130 nM for K562.

Per il saggio chinasi CDK1/ciclina B1, le reazioni chinasiche consistono in 100 ng di complesso GST-CDK1/ciclina B1 (umano) espresso da baculovirus, 1 μg di istone HI, 0,2 μCi di [γ-33P]ATP, 25 μM di ATP in 50 μL di tampone chinasico (50 mM Tris, pH 8.0, 10 mM MgCl2, 1 mM EGTA, 0.5 mM DTT). Per il saggio chinasi CDK2/ciclina E, le reazioni chinasiche consistono in 5 ng di complesso GST-CDK2/ciclina E (umano) espresso da baculovirus, 0,5 μg di proteina di fusione GST-RB (aminoacidi 776-928 della proteina del retinoblastoma), 0,2 μCi di [γ-33P]ATP, 25 μM di ATP in 50 μL di tampone chinasico (50 mM Hepes, pH 8.0, 10 mM MgCl2, 1 mM EGTA, 2 mM DTT). Per il saggio chinasi CDK4/ciclina D1, le reazioni chinasiche consistono in 150 ng di GST-CDK4/ciclina D1 (umano) espresso da baculovirus, 280 ng di Stag-ciclina D1, 0,5 μg di proteina di fusione GST-RB (aminoacidi 776-928 della proteina del retinoblastoma), 0,2 μCi di [γ-33P]ATP, 25 μM di ATP in 50 μL di tampone chinasico (50 mM Hepes, pH 8.0, 10 mM MgCl2, 1 mM EGTA, 2 mM DTT). Le reazioni vengono incubate per 45 minuti per CDK1 e CDK2, o 1 ora per CDK4 a 30 °C e interrotte con l'aggiunta di acido tricloroacetico (TCA) freddo a una concentrazione finale del 15%. I precipitati di TCA vengono raccolti su piastre unifilter GF/C utilizzando un raccoglitore universale Filtermate e i filtri vengono quantificati utilizzando un contatore a scintillazione liquida a 96 pozzetti TopCount. I valori di IC50 sono derivati mediante analisi di regressione non lineare e hanno un coefficiente di varianza = 16%. Per saggiare la sua attività su CDK6, viene stabilito un saggio di legame su filtro. Nel miscela di reazione vengono combinati i seguenti: 2 μL di CDK6 (0,7 mg/μL), 5 μL di istone H1 (6 mg/mL), 14 μL di tampone chinasico (60 mM β-glicerofosfato, 30 mM p-nitrofenil fosfato, 25 mM MOPS (pH 7.0), 5 mM EGTA, 15 mM MgCl2, 1 mM DTT, 0,1 mM Na-vanadato), 3 μL di concentrazioni crescenti di questo composto diluite in 50% DMSO e 6 μL di 33P-ATP (1 mCi/mL) in ATP non radioattivo a una concentrazione di 90 μM (concentrazione finale: 15 μM). Il saggio viene avviato con l'aggiunta di 33P-ATP. La reazione viene incubata per 20 minuti a 30°C. Un'aliquota di 25 μL del surnatante viene quindi spotta su carta di fosfocellulosa Whatman P81. I filtri vengono lavati 5 volte con una soluzione di acido fosforico all'1%. I filtri umidi vengono contati in presenza di 1 mL di fluido scintillatore. L'attività di Cdk9 viene misurata utilizzando 50 nM di Cdk9/ciclina T ricombinante in 50 mM HEPES pH 7.5, 10 mM MgCl2, 1 mM DTT, 3 μM Na3VO4, 150 μM di peptide CDT di RNA polimerasi e 80 μM di ATP. Il saggio Cdk7 viene eseguito nello stesso tampone utilizzando 37 nM di chinasi purificata in presenza di 200 μM di ATP e 10 μM di proteina legante la mielina come substrato. Administration of S1230 at 7.5 mg/kg for 7 days displays slight antitumor activity against P388 murine leukemia, resulting in %T/C value of 110, and active against the human A2780 ovarian carcinoma implanted sc in nude mice, producing 1.5 log cell kill (LCK). S1230 treatment at 1-2.5 mg/kg for 10 days significantly suppresses collagen-induced arthritis in mice in a dose-dependent manner, by inhibiting synovial hyperplasia and joint destruction, whereas serum concentrations of anti-collagen type II (CII) Abs and proliferative responses to CII are maintained. In the p21-intact Hct116 xenografts in nude mice, administration of CPT-11 (100 mg/kg) followed by S1230 (3 mg/kg) 7 and 16 hours later significantly inhibits tumor regression by 86% and 82%, respectively, displaying >2 fold inhibition compared with CPT-11 alone by 40 %. The combination produces ~30% complete response rate (CR) in contrast to CPT-11 alone where no CR is found.
