Mitoxantrone induce la frammentazione del DNA e la scissione proteolitica della poli(ADP-ribosio) polimerasi (PARP), un marcatore dell'attivazione delle caspasi, in tutti i pazienti studiati, dimostrando che l'effetto citotossico di mitoxantrone è dovuto all'induzione di apoptosi. Mitoxantrone attiva NFkappaB e stimola la degradazione di IkappaBalpha nella linea cellulare di leucemia promielocitica HL60 ma non nelle cellule varianti, le cellule HL60/MX2, che mancano dell'isoforma beta della topoisomerasi II ed esprimono un'isoforma alfa tronca che risulta in una distribuzione subcellulare alterata. Mitoxantrone inibisce la proliferazione di PBMCs attivati, linfociti B o linee cellulari T (TCLs) antigene-specifiche stimolate su cellule presentanti l'antigene (APCs) in modo dose-dipendente. Mitoxantrone induce l'apoptosi di PBMCs, monociti e DCs a basse concentrazioni, mentre dosi più alte causano la lisi cellulare. |

Mitoxantrone diminuisce transitoriamente il tasso di crescita degli xenotrapianti HID nei topi ma non influenza quello degli xenotrapianti PAC120. Mitoxantrone provoca la gravità delle lesioni cardiache e la nefropatia e la tossicità intestinale nei ratti spontaneamente ipertesi. Mitoxantrone e ferro(III) formano un forte complesso 2:1, in cui mitoxantrone può agire come un ligando tridentato. |
