AZD6482 inibisce anche PI3Kα, γ e δ, con IC50 da 80 nM a 1,09 μM, che sono significativamente inferiori rispetto al suo enantiomero (+)- (forma S). Questo composto è un agente antiaggregante piastrinico; blocca l'attivazione/aggregazione piastrinica e promuove la disaggregazione piastrinica in un saggio di aggregazione piastrinica lavata (WPA), con un valore IC50 di 6 nM. Inoltre, mirando a PI3Kβ, questa sostanza chimica inibisce specificamente la trombosi senza interferire con la normale emostasi. Pertanto, è usato come farmaco antitrombotico per la profilassi dei disturbi trombotici.

L'inibizione di PI3Kβ, PI3Kα, PI3Kγ e PI3Kδ viene valutata in un saggio di attività enzimatica basato su AlphaScreen utilizzando enzimi ricombinanti umani. Il saggio misura la conversione di PIP2 in PIP3 mediata da PI3K. Il PIP3 biotinilato, un dominio di omologia di pleckstrina (PH) marcato con GST e le due microsfere AlphaScreen formano un complesso che emette un segnale all'eccitazione laser a 680 nm. Il PIP3 formato nella reazione enzimatica compete con il PIP3 biotinilato per il legame al dominio PH, riducendo così il segnale con l'aumento del prodotto enzimatico. Questo composto è disciolto in DMSO e aggiunto a piastre a 384 pozzetti. PBKβ, PBKα, PBKγ o PBKδ viene aggiunto in un tampone Tris (50 mM Tris pH 7,6, 0,05% CHAPS, 5 mM DTT e 24 mM MgCl2) e lasciato pre-incubare con questa sostanza chimica per 20 minuti prima dell'aggiunta della soluzione di substrato contenente PIP2 e ATP. La reazione enzimatica viene interrotta dopo 20 minuti dall'aggiunta di una soluzione di arresto contenente EDTA e biotina-PIP3, seguita dall'aggiunta di una soluzione di rilevazione contenente GST-grpl PH e microsfere AlphaScreen. Le piastre vengono lasciate per un minimo di 5 ore al buio prima dell'analisi. Le concentrazioni finali di DMSO, ATP e PIP2 nel saggio sono, rispettivamente, 0,8%, 4 μM e 40 μM. I valori di IC50 sono calcolati secondo l'equazione, y = {a+[(b-a)/(l+(x/IC50)s)]}, dove y = % di inibizione; a = 0%; b = 100%; s = la pendenza della curva dose-risposta; x = la concentrazione di questo composto.
