Tanespimycin (17-AAG), un analogo della geldanamicina, mostra un'affinità di legame superiore a 100 volte per Hsp90 derivata da cellule tumorali con sovraespressione di HER-2 (BT474, N87, SKOV3 e SKBR3) o cellule di carcinoma mammario BT474 con valori di IC50 di 5-6 nM. Questo composto provoca la degradazione di HER2, HER3, Akt e del recettore degli androgeni (AR) sia mutante che wild-type, portando all'arresto della crescita G1 RB-dipendente di cellule di cancro alla prostata come LNCaP, LAPC-4, DU-145 e PC-3 con valori di IC50 di 25-45 nM. Oltre a indurre l'apoptosi delle cellule Ba/F3 trasformate con BCR-ABL wild-type con una IC50 di 5,2 μM, ha la capacità di indurre l'apoptosi delle cellule trasformate con i mutanti BCR-ABL T315I e E255K con valori di IC50 di 2,3 μM e 1,0 μM, rispettivamente, inducendo la degradazione sia della proteina BCR-ABL wild-type che dei mutanti. |

Proteina Hsp90 nativa purificata o lisati cellulari da cellule tumorali con sovraespressione di HER-2 (BT474, N87, SKOV3 e SKBR3) o cellule di carcinoma mammario BT474 in tampone di lisi (20 mM HEPES, pH 7.3, 1 mM EDTA, 5 mM MgCl2, 100 mM KCl) vengono incubati con varie concentrazioni di Tanespimycin (17-AAG) per 30 minuti a 4 °C, e poi incubati con biotina-GM legata a perline magnetiche di streptavidina BioMag per 1 ora a 4 °C. I tubi vengono posti su un supporto magnetico, e il supernatante non legato viene rimosso. Le perline magnetiche vengono lavate tre volte in tampone di lisi e riscaldate per 5 minuti a 95 °C in tampone campione SDS-PAGE. I campioni vengono analizzati su gel proteici SDS, e vengono eseguiti western blot usando gli anticorpi indicati. Le bande nei western blot vengono quantificate usando il Bio-rad Fluor-S MultiImager, e viene calcolata la percentuale di inibizione del legame di Hsp90 alla biotina-GM. L'IC50 riportata è la concentrazione di questo composto necessaria per causare un'inibizione della metà massima del legame.

La Tanespimycin (17-AAG) mostra un'affinità di legame significativamente più elevata per Hsp90 da xenotrapianti 3T3-src, B16 o CT26 in topi nudi con valori di IC50 di 8-35 nM rispetto a quella dei tessuti normali con valori di IC50 di 200-600 nM. L'amministrazione di questo composto (~50 mg/kg) provoca un significativo calo dell'espressione di AR, HER2, HER3 e Akt in modo dose-dipendente con un calo >50% alla dose di 50 mg/kg, con conseguente inibizione dose-dipendente della crescita di xenotrapianti di cancro alla prostata androgeno-dipendenti (CWR22) e indipendenti (CWR22R e CWRSA6) rispettivamente del 67%, 80% e 68% alla dose di 50 mg/kg. |

It can inhibit both HSP90α and HSP90β, but it is unclear how much they are affected respectively.
