BX-795 blocca efficacemente l'attività di PDK1 nelle cellule PC-3, come dimostrato dalla loro capacità di bloccare la fosforilazione di S6K1, Akt, PKCδ e GSK3β. Questo composto inibisce potentemente la crescita delle cellule tumorali su plastica con IC50 di 1,6, 1,4 e 1,9 μM per le cellule MDA-468, HCT-116 e MiaPaca, rispettivamente. In agar molle, mostra una maggiore inibizione della crescita con IC50 di 0,72 e 0,25 μM per le cellule MDA-468 e PC-3, rispettivamente. Inoltre, questa sostanza chimica, come inibitore di TBK1/IKKɛ, blocca l'attivazione di IRF3 mediata da TBK1 e IKKε e la produzione di IFN-β. Nelle risposte fisiologiche delle piastrine, questo composto produce un effetto inibitorio sull'aggregazione indotta da 2-MeSADP o da collagene, sulla secrezione di ATP e sulla generazione di trombossano. |

PDK1 viene saggiato in un saggio chinasico diretto e in un formato di saggio accoppiato che misura l'attivazione di AKT2 mediata da PDK1 e PtdIns-3,4-P2. Per il saggio accoppiato, la miscela finale del saggio (60 μL) conteneva: 15 mM MOPS, pH 7,2, 1 mg/mL di albumina sierica bovina, 18 mM di β-glicerolo fosfato, 0,7 mM di ditiotreitolo, 3 mM di EGTA, 10 mM di MgOAc, 7,5 μM di ATP, 0,2 μCi di [γ-33P]ATP, 7,5 μM di substrato peptidico biotinilato (biotin-ARRRDGGGAQPFRPRAATF), 0,5 μL di vescicole fosfolipidiche contenenti PtdIns-3,4-P2, 60 pg di PDK1 umano ricombinante purificato e 172 ng di AKT2 umano ricombinante purificato. Dopo incubazione per 2 ore a temperatura ambiente, il peptide marcato con biotina viene catturato da 10 μL della miscela del saggio su sfere SPA rivestite di streptavidina, e la formazione del prodotto viene misurata per scintillazione di prossimità in un contatore Wallac MicroBeta. Il prodotto formato è proporzionale al tempo di incubazione e alla quantità di PDK1 e AKT2 inattiva aggiunti. Questo composto viene aggiunto a livelli suboptimali in modo che il saggio possa rilevare sensatamente gli inibitori dell'attivazione di AKT2, così come gli inibitori diretti di PDK1 o AKT2. Per misurare direttamente l'attività di PDK1, la miscela finale del saggio (60 μL) conteneva 50 mM Tris-HCl, pH 7,5, 0,1 mM EGTA, 0,1 mM EDTA, 0,1% di β-mercaptoetanolo, 1 mg/mL di albumina sierica bovina, 10 mM di MgOAc, 10 μM di ATP, 0,2 μCi di [γ-33P]ATP, 7,5 μM di peptide substrato (H2N-ARRRGVTTKTFCGT) e 60 ng di PDK1 umano ricombinante purificato. Dopo 4 ore a temperatura ambiente, aggiungiamo 25 mM di EDTA e depositiamo una porzione della miscela di reazione su carta fosfocellulosa Whatman P81. La carta da filtro viene lavata tre volte con acido fosforico allo 0,75% e una volta con acetone. Dopo l'essiccazione, il peptide marcato legato al filtro viene quantificato utilizzando un fosforimager Fuji.
