Descrizione

Questo lattone steroideo, isolato da Withania somnifera, inibisce potentemente l'attivazione del NF-κB prevenendo l'attivazione indotta dal fattore di necrosi tumorale della IκB chinasi β tramite un meccanismo redox sensibile alla tioalchilazione. Si lega anche alla proteina del filamento intermedio (IF), la vimentina, con attività antitumorale e antiangiogenica.

La Withaferina A (WFA) inibisce potentemente l'attivazione del NFκB prevenendo l'attivazione indotta dal fattore di necrosi tumorale della IκB chinasi β tramite un meccanismo redox sensibile alla tioalchilazione. Si lega alla proteina del filamento intermedio (IF), la vimentina, modificando covalentemente il suo residuo di cisteina, presente nel dominio coiled-coil 2B α-elicoidale altamente conservato, e induce l'aggregazione dei filamenti di vimentina in vitro.

In vivo

La Withaferina A (WFA) mostra chiaramente caratteristiche antinfiammatorie in un modello murino di infiammazione acuta. La sua inibizione indotta della crescita capillare in un modello murino di neovascolarizzazione corneale è compromessa nei topi deficienti di vimentina.
