Descrizione OICR-9429 è un potente antagonista dell'interazione di WDR5 con le regioni peptidiche di MLL e Histone 3 e riduce la vitalità delle cellule di leucemia mieloide acuta in vitro. Si lega a WDR5 con alta affinità (KD = 93 ± 28 nM).

OICR-9429 si lega a WDR5 con alta affinità (Kd=93±28 nM) e interrompe competitivamente la sua interazione con un peptide MLL interagente con WDR5 (WIN) ad alta affinità (Kdisp=64±4 nM).
In vivo

OICR-9429, un potente inibitore di WDR5, ha soppresso la proliferazione, le metastasi e l'evasione immunitaria basata su PD-L1, aumentando al contempo l'apoptosi e la chemiosensibilità al cisplatino nel cancro alla vescica bloccando il complesso WDR5-MLL che media H3K4me3 nei geni bersaglio.
