Descrizione ML-792 è un inibitore potente e selettivo dell'enzima attivante SUMO (small ubiquitin-like modifier) (SAE). ML-792 inibisce SAE/SUMO1 e SAE/SUMO2 in saggi di scambio ATP-pirofosfato inorganico (PPi) con IC50 di 3 nM e 11 nM, rispettivamente.

ML-792 è un inibitore dell'enzima attivante SUMO (SAE) basato sul meccanismo con potenza nanomolare in saggi cellulari. ML-792 blocca selettivamente l'attività enzimatica di SAE e la SUMOylation totale, diminuendo così la proliferazione delle cellule tumorali. L'induzione dell'oncogene MYC aumenta l'effetto di vitalità mediato da ML-792 nelle cellule tumorali, indicando così una potenziale applicazione degli inibitori di SAE nel trattamento dei tumori amplificati MYC.

    HCT116 cells are plated (0.2 × 106 cells/well) and incubated overnight in six-well tissue culture dishes. To study ML-792-induced DNA damage, cells are then treated with DMSO or 0.5 μM ML-792 for 24 h. A 4-h treatment is used as a positive control for induction of DNA-strand breaks. To study how loss of SUMOylation contributes to repair of DNA damage, individual wells of HCT116 cells are treated with the following treatments: DMSO (6 h); 0.5 μM ML-792 (6 h).
