Descrizione MCC950 Sodium è un inibitore potente e selettivo di NLRP3 con IC50 di 7,5 nM in BMDM; ma non degli inflammasomi AIM2, NLRC4 o NLRP1.

In vitro

MCC950 inibisce l'attivazione canonica e non canonica di NLRP3 nelle cellule BMDM e HMDM a IC50 di 7,5 e 8,1 nM. MCC950 inibisce specificamente l'attivazione di NLRP3 ma non degli inflammasomi AIM2, NLRC4 o NLRP1.
In vivo

MCC950 riduce la produzione di interleuchina-1β (IL-1β) e attenua la gravità dell'encefalomielite autoimmune sperimentale (EAE). Inoltre, il trattamento con MCC950 salva la letalità neonatale in un modello murino di CAPS ed è attivo in campioni ex vivo di individui con sindrome di Muckle-Wells.
