Attività biologica

Descrizione KPT-185 è un inibitore selettivo di CRM1 che induce l'inibizione della crescita e l'apoptosi in un pannello di linee cellulari NHL con una IC50 mediana di ~25 nM.

In vitro KPT-185 inibisce significativamente la proliferazione delle cellule leucemiche con IC50 che vanno da 100 nM a 500 nM, e induce l'arresto del ciclo cellulare e l'apoptosi delle linee cellulari AML e dei blasti AML primari. Questo composto riduce il livello della proteina CRM1 e mostra un significativo accumulo nucleare delle proteine cargo di CRM1. Inoltre, inibisce la proliferazione e promuove l'apoptosi delle cellule di cancro al pancreas senza influenzare le cellule epiteliali duttali pancreatiche umane.

    Cells are seeded into 96-well plates and treated for 24, 48, and 72 hours with KPT-SINE at various concentrations ranging from 10 nM to 10 μM. Cell viability is evaluated using the cell proliferation reagent WST-1 according to the manufacturer's protocol. The absorbance of wells at 450 nm (reference wavelength, 650 nm) is measured with a microplate reader.
