Attività biologica

Descrizione Etravirine è un inibitore non nucleosidico della Reverse Transcriptase (NNRTI) utilizzato per il trattamento dell'HIV.
Target
Reverse transcriptase
In vitro Etravirine (TMC125) è altamente attivo contro l'HIV-1 wild-type con EC50 da 1,4 nM a 4,8 nM e mostra una certa attività contro l'HIV-2 con EC50 di 3,5 μM. TMC125 inibisce anche una serie di sottotipi di HIV-1 gruppo M e forme ricombinanti circolanti e un virus del gruppo O.
In vivo Etravirine ha un'elevata barriera genetica allo sviluppo di resistenza. Negli studi di fase IIb su pazienti con esperienza di trattamento, compresi quelli infettati da virus resistente agli NNRTI e agli inibitori della proteasi (PI), TMC125 è attivo contro l'HIV resistente agli NNRTI attualmente disponibili, con un profilo di tollerabilità simile a quello del gruppo di controllo.
