Descrizione A-1331852 è un potente e selettivo inibitore di BCL-XL con un valore di Ki inferiore a 0,01 nM per BCL-XL e 6 nM, 4 nM, 142 nM per Bcl-2, Bcl-W, MCL-1 rispettivamente. Può essere utile nel trattamento del cancro, delle malattie immunitarie e autoimmuni.

In vitro A-1331852 mostra una notevole potenza sia come agente singolo che in combinazione con i TKI nell'uccidere le cellule primarie CML CD34+. Inoltre, ha una notevole potenza nell'indurre Apoptosis in queste cellule a basse concentrazioni nanomolari già 1 ora dopo il trattamento.
In vivo A-1331852 dimostra efficacia antitumorale nel modello di xenotrapianto Molt-4, inducendo regressioni tumorali come agente singolo.

    Immunoprecipitation of BCL-XL is carried out in K562 cells, exposed to A-1331852 (100 nM) for 0-2 h, and the eluted complexes are immunoblotted for the indicated proteins. The input cell lysates and the immunodepleted supernatant (labeled as Flow-through) are immunoblotted to check the efficiency of the immunoprecipitation.
