CP-690550 è un inibitore orale specifico di JAK3, è 20-100 volte meno potente per JAK2 e JAK1 con IC50 di 20 nM e 112 nM, rispettivamente. Questo composto non ha una potente attività contro altre 30 chinasi (tutte le IC50 mediane > 3000 nM). Inibisce la proliferazione indotta da IL-2 con una potenza 30 volte superiore rispetto ai suoi effetti sulla proliferazione indotta da GM-CSF. Questa sostanza chimica inibisce efficacemente una reazione linfocitaria mista murina (MLR) (IC50 = 91 nM). Inibisce potentemente l'upregulation di CD23 indotta da IL-4 (IC50=57 nM) e l'espressione del complesso maggiore di istocompatibilità di classe II (MHCII) (IC50=71 nM) sulle cellule B murine. Una ricerca recente indica che una bassa dose di questo composto accelera l'insorgenza dell'encefalomielite autoimmune sperimentale potenziando la differenziazione Th17.

Un frammento che codifica il dominio catalitico di JAK3 umana (785aa a 1125aa, dominio catalitico JH1) viene amplificato mediante PCR dal cDNA a lunghezza intera e clonato nel sito EcoRI del vettore di trasferimento di baculovirus pVL1393. Il baculovirus ricombinante viene utilizzato per infettare le cellule Sf9 (Spodoptera frugipedra) e la proteina di fusione GSTJAK3 ricombinante viene isolata su sefarosio di glutatione. La proteina di fusione viene eluita con glutatione ridotto e conservata in un tampone contenente 50 mM Tris, pH 7,5, 10 mM DTT e 10% di glicerolo. L'attività della chinasi JAK3 viene misurata mediante ELISA come segue: Le piastre vengono rivestite durante la notte con un copolimero casuale di acido L-glutammico e tirosina (4:1) (100 ug/mL). Le piastre vengono lavate e la JAK3 JH1:GST ricombinante (100 ng/pozzetto) con o senza inibitori viene incubata a temperatura ambiente per 30 minuti, dopodiché viene aggiunto l'anticorpo anti-fosfotirosina PY20 coniugato con HRP (ICN) e sviluppato mediante TMB (3,3',5,5'-tetrametilbenzidina). Altre chinasi (Tabella 1) vengono prodotte in E. coli o in cellule di insetto, a seconda di ciò che risulta ottimale per la data chinasi. L'attività catalitica delle tirosina chinasi viene misurata utilizzando l'ELISA sopra menzionato, mentre le serina/treonina chinasi vengono saggiate utilizzando saggi enzimatici radioattivi.

L'EC50 (concentrazione del farmaco nel sangue alla quale il 50% dei topi manterrà il proprio innesto per >28 giorni) è di circa ~60 ng/mL. Questo composto previene il rigetto di reni allogenici in primati non umani (NHP, Macaca fascicularis) (MST di 62 e 83 giorni per i gruppi da 50 a 100 ng/ml e da 200 a 400 ng/ml, rispettivamente). I topi trattati cronicamente con questo composto (1,5-15 mg/kg/die) dimostrano alterazioni dose e tempo-dipendenti nei sottogruppi linfocitari quando esaminati mediante citometria a flusso. Il cambiamento più drammatico osservato è una riduzione del 96% nel numero di cellule spleniche NK1.1+TCRb dopo 21 giorni di trattamento. Le risposte di ipersensibilità di tipo ritardato (DTH) in topi sensibilizzati sono ridotte in modo dose-dipendente dopo il trattamento con questa sostanza chimica (1,87–30 mg/kg, s.c.). La sopravvivenza prolungata dei cuori neonatali Balb/c impiantati nel padiglione auricolare di topi C3H/HEN con MHC non corrispondente è osservata con questa monoterapia (10–30 mg/kg/die), ma migliorata in combinazione con la ciclosporina (10 mg/kg/die).
