AZD9291 mostra un'inibizione significativamente più potente della proliferazione in linee cellulari EGFR mutanti rispetto al wild-type in vitro.

Saggio di fosforilazione cellulare di EGFR
Le cellule vengono seminate (10000 cellule/pozzetto) in terreno di crescita in piastre Corning nere con fondo trasparente da 384 pozzetti e incubate a 37°C con 5% di CO2 durante la notte. Le cellule vengono dosate acusticamente utilizzando un Echo 555, con i composti diluiti in serie in DMSO al 100%. Le piastre vengono incubate per ulteriori 2 ore, quindi, dopo l'aspirazione del terreno, 40 μL di tampone di lisi 1x vengono aggiunti a ciascun pozzetto. Le piastre Greiner nere ad alto legame da 384 pozzetti vengono rivestite con anticorpo di cattura e quindi bloccate con 3% di BSA. Dopo la rimozione del blocco, 15 μL di lisato vengono trasferiti nelle piastre Greiner nere ad alto legame da 384 pozzetti e incubati per 2 ore. Dopo l'aspirazione e il lavaggio delle piastre con PBS, sono stati aggiunti 20 μL di anticorpo di rilevazione e incubati per 2 ore. Dopo l'aspirazione e il lavaggio delle piastre con PBS, 20 μL di substrato fluorogenico per la perossidasi QuantaBlu vengono aggiunti e incubati per 1 ora. 20 μL di soluzione di arresto QuantaBlu vengono aggiunti alle piastre e la fluorescenza viene letta su un lettore di piastre Envision utilizzando una lunghezza d'onda di eccitazione di 352 nm e una lunghezza d'onda di emissione di 460 nm. I dati ottenuti con ciascun composto vengono esportati in un pacchetto software adatto per eseguire l'analisi di adattamento della curva. Da questi dati, un valore di IC50 viene determinato calcolando la concentrazione del composto necessaria per ottenere un effetto del 50%.
In vivo

AZD9291 (5 mg/kg p.o.) provoca una profonda regressione dei tumori nei modelli tumorali EGFRm+ (PC9) ed EGFRm+/T790M (H1975) con una profonda inibizione della fosforilazione di EGFR e delle principali vie di segnalazione a valle come AKT ed ERK in vivo.
