L'estere metilico di NG-nitro-L-arginina (L-NAME; a 0,1-100 mM) provoca un'inibizione dipendente dalla concentrazione della NO sintasi endoteliale Ca2(+)-dipendente da aorte suine. Questo composto provoca una contrazione endotelio-dipendente e un'inibizione del rilassamento endotelio-dipendente indotto dall'acetilcolina (ACh) negli anelli aortici. In un'altra ricerca, la vitalità delle cellule rMC-1 o BREC in 25 mM di glucosio è significativamente inferiore rispetto a 5 mM di glucosio, e questa morte cellulare è inibita da questa sostanza chimica in entrambi i tipi di cellule.

L'ossidazione di L-NAME HCl (Nω-nitro-L-arginina metil estere cloridrato) è monitorata dalla conversione di [3H]- o [14C]-arginina in L-citrullina che separa L-citrullina da L-arginina mediante cromatografia Dowex 50x8-200 (Na). Le miscele di reazione tipiche (100 pL) contengono 50 mM HEPES, pH 7.0, 8 pM tetraidrobiopterina, 1 mM CaC12, 0.01 mg/mL calmodulina, 0.5 mM EDTA, 0.450 pM [14C]-arginina (30000 cpm) e 100-200 pM NADPH. L'ossidazione di NADPH a NADP+ catalizzata dalla cNOS è monitorata dalla riduzione dell'assorbanza a 340 nm con uno spettrofotometro Kontron 860 in un volume di 300 pL. Tutte le reazioni sono a 30 ℃ salvo diversa indicazione.

L-NAME HCl (0,03-300 mg kg-1, i.v.) induce un aumento dose-dipendente della pressione arteriosa sistemica media accompagnato da bradicardia. Questo composto (100 mg kg-1, i.v.) inibisce significativamente le risposte ipotensive ad ACh e bradichinina. L'aumento della pressione sanguigna e la bradicardia prodotti da questa sostanza chimica sono invertiti dalla L-arginina (30-100 mg kg-1, i.v.) in modo dose-dipendente.
