A concentrazioni del farmaco che riflettono la concentrazione plasmatica di picco dopo la somministrazione di Daunorubicin, il meccanismo primario è probabilmente attraverso l'interazione con la topoisomerase II, che può essere un evento scatenante primario per l'arresto della crescita e/o l'uccisione cellulare attraverso una via di segnalazione che porta all'apoptosis, almeno nelle cellule leucemiche e nei timociti. La struttura chinonica permette alla daunorubicina di agire come accettore di elettroni in reazioni mediate da enzimi ossidoriduttivi inclusi citocromo P450 reduttasi, NADH deidrogenasi e xantina ossidasi. A concentrazioni di Daunorubicin che superano circa 2-4 µM, la tossicità mediata dai radicali liberi e il cross-linking del DNA possono diventare evidenti. La Daunorubicin inibisce sia la sintesi del DNA che dell'RNA nelle cellule HeLa in un intervallo di concentrazione da 0,2 a 2 µM. La Daunorubicin inibisce la sintesi del DNA nelle cellule tumorali di ascite di Ehrlich in un intervallo di concentrazione di 4 µM. Il Daunorubicin innesca l'apoptosis a concentrazioni di 0,5 e 1 µM in cellule leucemiche umane HL-60 o U-937. La Daunorubicin stimola l'elevazione della ceramide e l'apoptosis nelle cellule P388 e U937 attraverso la sintesi de novo tramite l'attivazione dell'enzima ceramide sintasi. La Daunorubicin aumenta in modo dose-dipendente l'esposizione alla fosfatidilserina e la conseguente attività procoagulante delle cellule endoteliali della vena ombelicale umana. La Daunorubicin (0,2 mM) aumenta significativamente il rilascio di microparticelle endoteliali che sono altamente procoagulanti nelle cellule endoteliali della vena ombelicale umana. |
