Il solfato di vincristina inibisce l'aggiunta netta di dimeri di tubulina alle estremità di assemblaggio dei microtubuli allo stato stazionario con Ki di 85 nM. A basse concentrazioni, questo composto stabilizza l'apparato del fuso, con conseguente fallimento della segregazione dei cromosomi che porta all'arresto metafasico e all'inibizione della mitosi. A concentrazioni più elevate, può interrompere e indurre la depolimerizzazione totale dei microtubuli. Questa sostanza chimica induce l'apoptosi nelle cellule tumorali e inibisce la proliferazione delle cellule SH-SY5Y con IC50 di 0,1 μM. Induce l'arresto mitotico e promuove l'espressione di caspasi-3 e -9 e ciclina B, diminuendo l'espressione di ciclina D. La sua neurotossicità indotta è causata dall'interferenza con la funzione dei microtubuli, che si traduce nel blocco del trasporto assonale e quindi nella degenerazione assonale.

Il solfato di vincristina (3 mg/kg) somministrato con una singola iniezione i.p. a topi portatori di xenotrapianti sottocutanei bilaterali Rh12 o Rh18, induce un ritardo medio della crescita di >120 e >52 giorni, e frazioni di ripopolazione rispettivamente dello 0,06% e del 5%. Questo composto agisce sui tumori del colon 38 sottocutanei nei topi attraverso effetti vascolari mediati dalle cellule ospiti, nonché tramite citotossicità diretta mediata dalla tubulina. Esso (5 mg/kg) riduce il flusso sanguigno dei tumori di quasi il 75%.
