AZD8055 mostra una bassa attività (circa 1.000 volte) contro tutte le isoforme di PI3K (α, β, γ, δ) e altri membri della famiglia delle chinasi simili a PI3K (ATM e DNA-PK). Questo composto inibisce la fosforilazione di mTORC1 (p70S6K e 4E-BP1) così come la fosforilazione di mTORC2 (Akt) e delle proteine a valle. I siti di fosforilazione T37/46 su 4E-BP1, resistenti alla rapamicina, possono essere completamente inibiti da questo composto, con conseguente significativa inibizione della traduzione cap-dipendente. Inibisce potentemente la proliferazione nelle cellule U87MG, A549 e H838 con IC50 rispettivamente di 53, 50 e 20 nM. Questa sostanza chimica induce anche l'autofagia e l'aumento dei livelli di LC3-II nelle cellule H838 e A549. Diminuisce la proliferazione delle cellule blastiche AML e la progressione del ciclo cellulare, riduce la crescita clonogenica dei progenitori leucemici e induce l'apoptosi dipendente dalla caspasi nelle cellule leucemiche ma non nelle normali cellule CD34+ immature. Questo composto indica un'attività inibitoria contro le linee cellulari del programma di test preclinici pediatrici (PPTP) con un IC50 di 24,7 nM e induce differenze significative nella distribuzione dell'EFS.

Determinazioni dell'inibizione di mTOR basate su cellule

È stato sviluppato un saggio di screening ad alto rendimento basato su cellule utilizzando cellule MDA-MB-468 per rilevare l'attività di mTORC1 e mTORC2. Le cellule vengono esposte per 2 ore a concentrazioni crescenti di AZD8055. Al termine del periodo di incubazione, le cellule vengono fissate, lavate e sondate con anticorpi contro S473 pAkt o contro S235/236 S6 fosforilato (pS6). I livelli di fosforilazione vengono valutati utilizzando un citometro a scansione laser Acumen.

AZD8055 inibisce pS6 e pAkt in xenotrapianti U87MG e A549 a 2,5/10 mg/kg, il che porta all'inibizione della crescita tumorale. Questo composto mostra una significativa attività antitumorale in molti xenotrapianti, inclusi U87MG, BT474c, A549, Calu-3, LoVo, SW620, PC3 e MES-SA a una dose di 10/20 mg/kg. Induce una riduzione di circa il 40% del volume tumorale, accompagnata dall'ablazione della fosforilazione delle chinasi proteiche Akt, S6K e SGK. La somministrazione di questo composto chimico (5 mg/kg, Bid) e SAHA (100 mg/kg/die) si traduce in una completa inibizione della crescita tumorale in xenotrapianti PTEN+/-LKB1+/hypo senza effetti collaterali sui topi attraverso l'inibizione della segnalazione di mTORC1 e mTORC2.
