Biologische activiteit

Beschrijving Tacrolimus (FK506) is een 23-koppig macrolide lacton, het vermindert peptidyl-prolyl isomerase activiteit in T-cellen door te binden aan het immunofiline FKBP12 (FK506 binding protein) waardoor een nieuw complex ontstaat. Tacrolimus remt ook de phosphatase activiteit van calcineurin. Tacrolimus induceert vasculaire endotheliale autophagy.

In vitro

Tacrolimus (FK506) en cyclosporine A blokkeren translocatie van de cytoplasmatische component zonder de synthese van de nucleaire subeenheid in T-lymfocyten te beïnvloeden. Deze verbinding voorkomt T-celproliferatie door een Ca(2+)-afhankelijke gebeurtenis te remmen die nodig is voor de inductie van interleukine-2 transcriptie. Het bindt aan verschillende families van intracellulaire eiwitten (immunofilines) genaamd cyclofilines en FK 506-binding proteins (FKBPs). Tacrolimus remt specifiek cellulaire calcineurin bij medicijnconcentraties die de interleukine 2-productie in geactiveerde T-cellen remmen. Het en CsA oefenen bijna identieke biologische effecten uit in cellen door dezelfde subset van vroege calcium-geassocieerde gebeurtenissen te remmen die betrokken zijn bij lymfokine-expressie, apoptose en degranulatie. Deze verbinding bindt aan een familie van intracellulaire receptoren genaamd de FK-506 binding proteins (FKBPs).

In Vivo

Tacrolimus (FK506) resulteert in een toename van de terugtrekdrempel van de poot en staart, zoals blijkt uit gedragsmatige pijnbeoordeling bij ratten tegen hyperalgesische en allodynische stimuli. Het leidt ook tot een afname van de serum nitraat- en thiobarbituurzuur-reactieve stof (TBARS) -niveaus, samen met een vermindering van weefselmyeloperoxidase (MPO) en totale calciumniveaus, terwijl een stijging van de weefsel gereduceerde glutathionniveaus bij ratten optreedt. Deze verbinding verbetert de toename van neuronaal oedeem en axonale degeneratie bij ratten met ischemie-reperfusie (I/R).
