 0.300mg/ml (0.64mM) Taking the 1 mL working solution as an example, add 50 μL of 6 mg/ml clarified 5%TFA stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. The mixed solution should be used immediately for optimal results.

Beschrijving Dabigatran is een krachtige non-peptide thrombineremmer met een IC50 van 9,3 nM in een celvrije test.

In vitro BIBR 953 is een zeer krachtig anticoagulans. Deze verbinding toont aan dat de terminale fenyl kan worden vervangen door de meer hydrofiele 2-pyridylgroep zonder aanzienlijk verlies van activiteit. Het remt Thrombin, plasmine, factor Xa, trypsine, tPA en geactiveerd proteïne C met Ki van respectievelijk 4,5 nM, 1,7 μM, 3,8 μM, 50 nM, 45 μM en 20 μM. Deze chemische stof remt specifiek en reversibel Thrombin.
In Vivo BIBR 953 vertoont het meest gunstige activiteitsprofiel na i.v. toediening aan ratten. De biologische beschikbaarheid van deze verbinding na p.o. toediening van dabigatran etexilaat is 7,2%. Het wordt voornamelijk uitgescheiden in de feces na p.o. behandeling en in de urine na i.v. behandeling. De gemiddelde terminale halfwaardetijd van deze chemische stof is ongeveer 8 uur. De acylglucuroniden zijn goed voor respectievelijk 0,4% en 4% van de dosis in de urine na p.o. en i.v. dosering.
Functies Dabigatran is een omkeerbare, competitieve, directe Thrombin-remmer.

NPP was spiked with increasing concentrations of dabigatran and divided in two portions. One portion was left unaltered, while the other portion was treated with DOAC Stop (1 mL per tablet).
